摘要:目的 快速检测啤酒花浸膏、溴酸钾和焦磷酸钠等3种食品添加剂对V79细胞DNA的损伤作用。方法采用V79细胞培养和单细胞凝胶电泳技术,借鉴单细胞凝胶电泳技术(SCGE)图像分析系统IMI1.0对彗星图像进行分析,观察指标有尾长、尾重心、olive尾矩、尾惯量、尾分布矩、尾长/头长比、尾DNA含量(%)等7个计量指标。结果实验结果表明,溴酸钾和焦磷酸钠的剂量分别在75~300 μg/ml和25~100 μmol/ml范围内,均对V79细胞DNA有明显的损伤作用,存在剂量-反应关系;啤酒花浸膏的剂量在25~100μg/ml范围内,不引起V79细胞DNA的损伤作用。结论本研究方法与传统的彗星实验相比,具有分析指标多,操作方便等优点,可应用于快速检测食品添加剂诱变性的筛选试验。

Assay study on damnification for DNA of V79 cell caused by foodaddition agent by single cell gel electrophoresis

Abstract: Objective To study rapid test method ofthe damnification for the DNA of V79 cell by European hops extract、potassium bromate and sodium pyrophosphate. Methods The single cell gel electrophoresis assay and V79cell culture technic were used, the SCGE Image analyse systemIMI1.0 was adopted to analyze comet image for reference. The indexincluded length, barycenter,square、inertia、moment ofdistribution、ratio of length/head length and content of DNA(%). Results The result of the study show that : It’llcause distinctness damnification for the DNA of V79 cell whenthe dosage of potassium bromate and sodium pyrophosphate werein the range of 75~300 μg/ml and 25~100 μmol/ml respectively, andit exist the relationship of dose-reaction. Meanwhile, when theEuropean hops extract was in the range of 25~100 μg/ml,it’ll notcause damnification for DNA of V79 cell. Conclusion With the traditional comet experiment,the study has theexcellence such as more analyse index, convenient operate aswell.The study could apply to the filtration experiment forthe rapid test of themutagenicity of food additives.
