摘要:目的 对上海浦东机场蝇类携带新病原菌进行分离鉴定。方法将上海浦东机场口岸捕获的蝇体细菌分离培养至单克隆,使用Vitek2 Compact进行初步鉴定,扩增16SrRNA基因,进行序列比对。使用Mega5,Neighbour-joining方法,构建分子系统发育树。结果从大头金蝇体表分离出一株菌,疑似造成暴发性脓毒症的污蝇解壳杆菌(Wohlfahrtiimonaschitiniclastica)。进化树分析表明为同种菌,16S rRNA相似性为98.66%。结论 从大头金蝇体表成功分离到W.chitiniclastica菌的新记录。

Isolation of a new vector-borne pathogenic bacteria and
molecular identification

Abstract: Objective To identificationa new fly-borne pathogenic bacteria. Methods In vitro and in vivo bacteria of fly werecultured respectively. Monoclonal strain was identified byVitek2 Compact firstly and then 16S rRNA sequence was amplified andanalyzed. Phylogenetic tree was constructed by Mega5 software,Neighbour-joining method. Results A bacteria wasisolated from Chrysomyia megacephala, its 16S rRNA sequence sharedsimilarity of 98.86% with Wohlfahrtiimonas chitiniclastica, whichcaused fulminant sepsis in homeless patients. The phylogeneticanalysis indicated that the strain was the same species withWohlfahrtiimonas chitiniclastica. Conclusion A new record of W. chitiniclastica strainwas isolated from Chrysomyia megacephala collected from theport.
