摘要:目的探索对人致病的副溶血性弧菌的检测方法,评价5种检测方法对副溶血性孤菌致病性测定的有效性。方法采用耐热溶血毒素tdh基因和耐热直接溶血毒素相关毒素trh基因实时荧光定量PCR检测、并与神奈川溶血试验、小鼠腹腔注射和灌胃法对比,分别检测分离自腹泻者和自然环境中副溶血性弧菌标本。结果两类来源的副溶血性弧菌在tdh基因的检出率具有统计学意义(χ2=169.5,P<0.01),在神奈川溶血试验中差异有统计学意义(χ2 = 29.3,P <0.01),在trh基因、小鼠腹腔注射以及灌胃检测无统计学意义。结论tdh基因实时荧光定量PCR法是快速筛查致病性副溶血性弧菌的最好方法。

Abstract: Objective To compare five kinds of laboratory methods for the effectivenessof detecting pathogenic Vibrio parahaemolyticus strains. Methods Real time tdh/trh florescent quantitativepolymerase chain reaction (PCR), Kanagawa hemolytic test and mousetoxicity model by celiac injecting or fill in stomach wereused to compare their effectiveness on detection of pathogenicVibrio parahaemolyticus strains which were isolated either fromhospital diarrhea patients or natural environment. Results The detection rates of the presence of tdh genefrom two V. parahaemolyticus isolate resources were significantlydifferent from each other (χ2=169.5,P < 0.01). The positiverates of Kanagawa hemolytic tests were also different between twoV. parahaemolyticus populations(χ2=29.3,P < 0.01). However,differences were not significantly observed for the detectionmethods of trh PCR or mouse toxicity test between two V.parahaemolyticus populations. Conclusion The tdh PCRtest is more preferred for the identification of those pathogenicisolates from normal V. parahaemolyticus population.
