摘要:目的研究适合口岸媒介生物监测的蚊虫样本保存方法,并对蚊虫RNA保护效果进行评价。方法采用普通冰箱、低温冰箱、液氮、RNALater等保存方式,对蚊虫进行1周、1个月、6个月保存,提取蚊虫RNA,比较这些保存方式对蚊虫RNA保护效果。结果RNALater与液氮对蚊样本均具有良好的保存效果,在1周、1个月、6个月内,蚊虫RNA均未见明显降解;低温冰箱在1周、1个月内对蚊样本有较好的保存效果,蚊虫RNA未见明显降解,但6个月后,蚊虫RNA降解明显;常温冰箱(-20℃)保存效果不佳,仅在1周之内对蚊虫核酸有一定的保护效果,1个月后,RNA发生明显降解。结论RNALater、液氮和低温冰箱在1个月内对蚊虫RNA均具有较好的保存效果,但RNALater具有保存时效长、使用方便等优点,更适用口岸蚊虫监测中样本的保存和运输。

The evaluation of the protective effect ofdifferent
preservation methods to mosquito RNA

Abstract: Objective Searching for preservation methods of mosquito samples forvector-monitoring at ports,and to evaluate protective effect ofthese preservation methods. Methods Theordinary refrigerator, low temperature refrigeration, liquidnitrogen and RNALater preservation methods were used, meanwhile,the mosquitoes were preserved for a week, a month and six months,and then the mosquito RNA was extracted. Results The RNALater and nitrogen had the bestprotective effect for mosquito samples, the mosquitoes RNA were noobvious degradation within a week, a month and six months; lowtemperature refrigerator had better protective effect, mosquitoesRNA has no significant degradation in a week and one month, butafter six months, mosquito RNA degradation significantly; theordinary refrigerator (-20 ℃) poorly preserved, it only had aprotective effect for mosquitoes RNA within a week, more than aweek later, RNA was obvious degraded. Conclusions RNALater, low-temperature refrigerator and liquid nitrogen all hadbetter protective effect for mosquito samples in one month, butRNALater had advantage of a long time protection effect andconvenient, it is more suitable for mosquito sample preservationand transportation at ports.
