大榭港区啮齿动物携带汉坦病毒基因分型和序列分析

摘要 〔目的〕 研究大榭港区啮齿动物中汉坦病毒的感染流行情况以及病毒基因分型。〔方法〕采用夹夜法捕捉鼠类,直接免疫法(DIF)检测鼠肺中的汉坦病毒抗原;巢式RT-PCR法扩增阳性标本中核苷酸片段并测序,构建系统发生树进行分型与系统发生分析。〔结果〕共捕获鼠253只,经鉴定隶属2目2科5属7种。经检测,其中2份样本为汉坦病毒阳性,阳性检出率为0.79%,宿主均为社鼠,核苷酸序列同源性及系统发生树分析表明分型均为汉滩型(HTN型)。〔结论〕大榭港区啮齿动物汉坦病毒的阳性检出率比较低,以HTN型为主,宿主为社鼠。

Genotyping and DNA Sequenece Analysis ofHantaviruses at Daxie Port Hu Qun, Tong Shumei, GuoLiping. Daxie Exit-Entry Inspection and Quarantine Bureau, Ningbo315812,China
[Abstract] Objective To investigate the Hantanvirus infection and their genotype atDaxie Port. Method Rodents were collected fromthe port areas to detect antigen of Hantavirus in rat lungs bydirect immunofluorescence (DIF). The genotypes of viruses wereidentified by nested RT-PCR and sequenced. The obtained sequenceswere analyzed by phylogenetic tree software. Result All the captured rodent added up to 253. Therewere 7 species, 5 genera, 2 families, 2 order. 2 of 253 rats wereHantavirus-positive, the positive rate was 0.79%. The Hantaviruswas carried by Rattus niviventer. Homological and phylogeneticanalysis showed the hantavirus at Daxie Port belonging to a geneticsubtype HTN. Conclusion Data indicated thatthe rate of carring virus is row and the main genetic subtypeof Hantavirus is HTN and carried by Rattus niviventer.
