摘要 〔目的〕 建立胶体金免疫层析技术快速定量检测单增李斯特菌方法。〔方法〕利用胶体金标记和双抗体夹心免疫层析技术,建立单增李斯特菌的快速检测方法,评价其特异性和敏感性,并拟合标准曲线进行定量检测;在牛奶、橙汁、果冻等即食食品样品中添加单增李斯特菌模拟污染样品,评价该方法对半固体、液体等即食食品、可疑生物恐怖样品的检测能力。〔结果〕该法可在10 min内完成定性和半定量检测,定量检测灵敏度为3.5 × 103 cfu/ml,线性范围3.5 × 105 ~ 3.5× 108 cfu/ml,回收率在99%~101.7%之间。〔结论〕建立的检测单增李斯特菌的胶体金免疫层析方法,能快速、灵敏、特异、准确地对样品中的单增李斯特菌进行定性、定量检测。

[Abstract] Objective To develop a rapid and quantitative detection method on Listeriamonocytogenes using colloid gold immunochromatographicmethod. Methods The rapid detection method wasestablished with double-antibody sandwich assay, and thespecificity and sensitivity of the method was then evaluated.Quantification was realized by constructing a standard curve, Milkpowder,milk solution and ham sausage, added with ricn were testedto evaluate the feasibility on solid, semi-solid, liquid foodsamples and doubtful bioterrorism samples by this method. Results This established method could beaccomplished qualitatively as well as semi-quantitative detectionin 15 minutes, the sensitivity was up to 3.5×103 cfu/ml, the linearrange was from 3.5×105cfu/ml to 3.5×108cfu/ml, the recovery ratewas 99%~101.7%. This method showed good specificity andsensitivity, which could detect samples directly. Conclusion The colloidal gold immunochromatographic test, which could detectthe abrin in samples rapidly, specifically, sensitively andaccurately, was able to realize quantification, and is suitable forfield detection.
